I. Background
CD40 是腫瘤壞死因子受體超家族（TNFRSF）成員，作為免疫調節的關鍵分子，廣泛參與 B 細胞活化、抗原呈遞細胞（APC）成熟及 T 細胞免疫應答。其配體 CD40L（CD154）主要表達于活化的 T 細胞，通過 CD40-CD40L 互作橋接先天性與適應性免疫，是治療、自身免疫病及感染性疾病的熱門靶點。 CD40 信號轉導主要利用 TNF 受體相關因子的銜接蛋白，導致經典和非經典 NFκB 通路、MAPK、PI3K 和 PLCγ的激活。它們的激活導致特征性下游效應，包括轉錄激活、細胞骨架重排和細胞存活。
